Xenograft Models and In Vivo Tumor Imaging

Subcutaneous tumor xenograft models for testing candidate drug efficacy.

Standard study involves the following:
- culturing of cell line (may be also customer-provided),
- engraftment of cells subcutaneously to immunodeficient nu/nu mice,
- measurement of animal weight and tumor volume (15 timepoints, e.g. with prostate cancer PC3 cells 3 times per week),
- administration of test compound 15 times (e.g. 5 times per week for 3 weeks) to mice after randomisation,
- detailed final report including full statistical analysis.

Optional:
- monitoring can be prolonged,
- blood and tumor sample collection and shipment,
- in vivo imaging of tumor cells that express luminescent and fluorescent reporters (see also below).


Stable expression of luminescent and fluorescent marker proteins in cultured cells for in vivo imaging.

Quantitative imaging of cells expressing suitable fluorescent markers or firefly luciferase in mouse xenograft efficacy studies is also possible. For this, we use lentiviral transduction to stably label different cell lines with reporters (e.g. melanoma B16F10-luc, PC3-luc, PC3-GFP, see example: panels represent growth of a PC3-GFP tumor).
